Principle Specimens are plated on media that support the growth of fungi. Since hair, skin, and nail specimens commonly contain contaminating saprophytic fungi and bacteria, they are also plated on media containing cycloheximide and chloramphenicol (Mycobiotic or Mycosel) in order to recover the slower growing dermatophytes.
Container type sterile, screw-capped container
Collection Instructions 1. Skin: Clean the site with 70% ethanol and allow the alcohol to evaporate. Collect skin scrapings from the outer margin of the lesion in a sterile, screw-cap container.

2. Nails: Clean the site with 70% ethanol and allow the alcohol to evaporate. Collect nail scrapings from soft keratin beneath the nail plate and from the underside of the nail and place in a sterile, screw-cap container.

3. Hair: Use a Wood's Lamp (filtered ultraviolet) to find fluorescent hairs to be plucked or scrape scalp and send segments of hairs near the scalp from affected area. Place in a sterile, screw-cap container.
